We are looking for a motivated PhD student to work on the redox regulation of lysosome using a yeast model. This PhD thesis will be under de direction of Prof. Cédric DELPORTE (RD3-PBDD unit and APFP, Faculty of Pharmacy) and co-direction of Prof. Bruno ANDRÉ (Molecular Physiology of the Cell lab, Faculty of Sciences). The research will be mainly conducted at the Faculty of Pharmacy located in Brussels on the Plaine campus of ULB in Ixelles. Furthermore, some experiments will also be performed in the laboratory of Molecular Physiology of the Cell, Faculty of Sciences, in Gosselies.
The research project entitled “Yeast as a model to investigate the mechanisms of lysosome redox control” aims at studying the mechanisms regulating the redox state of the yeast vacuole used as a model of the lysosome. These mechanisms remain unknown and their dysfunction may accelerate ageing and cause diseases. In this part of the project, we will use liquid chromatography and mass spectrometry methods to identify the main redox-active compounds present in the sap of yeast vacuoles. We will attempt to identify and characterize the membrane transporters that mediate transport of these compounds into the vacuole and we will study their contribution to vacuolar redox homeostasis.
As member of APFP, the proposed PhD position will mainly focus on analytical developments in liquid chromatography coupled to tandem mass spectrometry (LC-MS(/MS)) to identify and quantify compounds in redox regulation in yeast vacuole. The job will be dedicated to
- Analytical development in LC-MS(MS) for detecting and quantifying redox compound including: cysteine, cystine, homocysteine, homocysteine, glutathione, glutathione disulfide, D-erythroascorbic acid, monodehydroerythroascorbic acid … from yeast vacuole (70 %)
- Sampling of yeast vacuoles under different conditions (20 %)
- Helping in the model development for understanding redox control in the vacuole (10 %)
Profile
Master in Pharmacy, Biochemistry, Chemistry, Analytical Chemistry, Biology or Bioengineering is mandatory. The candidate should have a good level of success during her/his studies and she/he should be distinguishing herself/himself during her/his master.
Candidates are expected to show
- Excellent organizational and time management skills
- Good skills in spoken and written English.
- High motivation, flexibility as well as innovate and creative thinking
Candidates with experience in the following items will have an advantage
- Experience in mass spectrometry analysis
- Experience in liquid chromatography coupled to mass spectrometry
- Experience in sample treatment (SPE, liquid/liquid extraction)
