PhD Position Available in

Cyclic LipoPeptide Structure and Mode of Action

as part of the UGent Concerted Research Action MEMCLiP

and the FWO Excellence of Science RhizoCLiP program

A PhD position is available within the NMR and Structure Analysis (Prof. José Martins) research group embedded in the Department of Organic and Macromolecular Chemistry and involved in the NMR Centre of Expertise.

CLiPs or cyclic lipodepsipeptides are secondary metabolites produced by Pseudomonas and Bacillus species, among others. They contribute to a wide range of biological effects in the context of plant-pathogen interactions but also present antimicrobial and anticancer properties (Geudens et al, 2018). In spite of clear application potential in this respect, a considerable gap with respect to understanding the mode of action still needs to be filled (see editorial). Two major research efforts, both within UGent and within a interuniversity context, aim to raise our fundamental physical, chemical and biological understanding of these compound. The ultimate focus of these research efforts is to engage in future valorization programs. The position will involve the production of CLiPs from bacterial growth, isolation and purification of natural and isotopically labeled CLiPs. A broad range of solution NMR spectroscopic methods will be applied in membrane mimicking conditions (micelles and isotropic bicelles) for the investigation of structure, location, orientation, dynamics and membrane interactions of these CLiPs (see for instance Geudens et al, 2019). Studies will be complemented by molecular dynamics investigations of the peptide-membrane interactions.

The ideal candidate has a Masters or equivalent degree in Chemistry, Biochemistry, Biophysics or a suitably related area. She/he is acquainted with the physical-theoretical background of NMR spectroscopy and already has hands-on experience with NMR equipment, as evident for instance from their Master’s Degree.

SELECTION CRITERIA

Essential : A passion for NMR spectroscopy

• Experience with 1D and 2D NMR techniques for biomolecular NMR investigations, preferably peptides or proteins

• Experience with setting up and performing NMR measurements

• Well-developed laboratory and problem-solving skills

• Ability to work independently within a collaborative environment

• Good literature mining skills

• Ability to write research manuscripts

• Good written and verbal communication skills in English

Of added value

• Experience with mass spectrometry

• Experience with HPLC analysis and purification

• Experience with investigations of dynamics through relaxation NMR measurements

• Experience with other biophysical methods
